چکیده
Silymarin (SM) is the active component of milk thistle (MT) that is a flavonoid complex consisting of silibinin, silychristin, and silydianin. Silibinin (also known as silybin) is the major active component of the silymarin complex and is credited with giving MT its beneficial properties. Acting as a phyotonutrient silybin also provides a variety of benefits including anti-inflammatory, anti-carcinogenic and cardiovascular benefits. SM may involve suppression of NF-kappa B, a nuclear transcription factor, which regulates the expression of various genes involved in inflammation, cytoprotection, and carcinogenesis. SM blocked TNF-induced activation of NF-kappa B in a doseand time-dependent manner. This effect was mediated through inhibition of phosphorylation and degradation of Iota kappa B alpha, an inhibitor of NF-kappa B. SM significantly reduces apoptosis, skin oedema, and depletion of catalase activity and induction of cyclooxygenase activity. This provides protection against burn-induced oxidative skin injury and photocarcinogenesis. Furthermore, SM significantly decreased skin cancer number and size in a validated model of tumor promotion. These findings are supported by other studies that show similar effects for Silibinin including protects against UV lightinduced DNA damage and cancer cell growth. Moreover, silibinin enhances the powerful tumor suppressor gene p53, a genetic factor that protects against cancer. Silibinin acts by other mechanisms to prevent UV lightinduced skin cancer. In fact, some findings suggest that silibinin can help to repair DNA damage caused by previous exposure to UV light.

Activation of Wnt signaling reduces high-glucose mediated damages on skin fibroblast cells
Objective(s): High-glucose (HG) stress, a mimic of diabetes mellitus (DM) in culture cells, alters expression of a large number of genes including Wnt and NF-κB signaling-related genes; however, the role of Wnt signaling during HG-mediated fibroblast damage and the relationship between Wnt and NF-κB signaling have not been understood. In this study, we aimed to investigate the ffects of Wnt sig...
متن کاملThe Role of Cyclooxygenase-2 in Signaling Pathways Promoting Colorectal Cancer
Colorectal cancer is one of the most common cancers in the world. Various factors are involved in the development and progression of this disease. One of these agents is cyclooxygenase-2 (COX-2). COX-2 is a product of the PTGS2 gene and converts free arachidonic acid to prostaglandins. COX-2 is not naturally expressed in most normal cells. Noticeably, the increased expression of COX-2 has been ...
متن کاملThe Canonical Wnt Signaling (Wnt/β-Catenin Pathway): A Potential Target for Cancer Prevention and Therapy
Precise regulation of signal transduction pathways is crucial for normal animal development and for maintaining cellular and tissue homeostasis in adults. The Wnt/Frizzled-mediated signaling includes canonical and non-canonical signal transduction pathways. Upregulation or downregulation of the canonical Wnt-signaling (or the Wnt/β-Catenin signal transduction) leads to a variety of human diseas...
متن کاملThe Role of Microgravity in Cancer: A Dual-edge Sword
Since human beings could travel beyond the earth atmosphere, scientists started to investigate the effect of microgravity on human cells. Microgravity has different effects on normal and cancer cells, but the related mechanisms are not well-known till now. The aim of the present review is to focus on the consequences of exposing the cancer cells to reduced gravity. Some cancer cells organize th...

Thyroid cancer is one of the most common malignancies of endocrine glands, causing carcinomas, such as papillary, follicular, medullary, and anaplastic thyroid carcinomas. Due to the significance of thyroid carcinomas, identification of the main signaling pathways and the affecting mutations has been considered by researchers. Further studies on the dysregulation of oncogenes in signaling path...
